【Abstract】 Objective To explore the silencing effect of the constructed mice′s matrix metalloproteinase (MMP)9 RNA interference (RNAi) lentivirus vector in vitro. Methods 293T and NIH3T3 cells were cocultured by MMP9 overexpressed clone plasmid and RNAi lentivirus vector. Real time PCR and Western blot were adopted to observe the downregulation of MMP9 mRNA expression in 293T and NIH3T3 cells by RNAi. Results The constructed MMP9 RNAi lentivirus vector significantly decreased MMP9 expression in 293T and NIH3T3 cells. Conclusions RNAi lentivirus vector inhibiting MMP9 expression is successfully constructed, laying foundation for the research in vivo.

Abstract:WRKY transcription factor family can help improve plant stress tolerance, which widely exist in various plants. After TaWRKY gene was silenced by VIGS method, it was found that the proportion of succeed Bgt inoculation increased, and the percentage of abnormal appressoria declined, such as papilla. The results indicated that TaWRKY gene played an important role in wheat- Bgt interaction.

小麥白粉病是由布氏白粉菌(Blumeria graminis f. sp. Tritici)侵染所引起的真菌感染性病害,如遇高溫多濕天氣病害流行,會使小麥嚴(yán)重受害,導(dǎo)致減產(chǎn)13%~34%[1]。 因此,科學(xué)家一方面通過抗病育種,不斷培育新的抗病小麥品種來抵御病害,另一方面通過深入的抗病分子機(jī)理研究,克隆抗病相關(guān)基因、弄清抗病信號通路以及基因工程等技術(shù)手段,以達(dá)到抗病分子育種的目的。
